The balance between survival and death signalling\nplays a critical r ole in determining the outcome of an immune\nresponse and the subsequent s urvival and maintenance of memory T\ncells. TNFR-associated factors (TRAFs ) are a family of adaptor\nproteins involved in linking TNFR family member s to downstream\nsignalling pathways. TRAF1 is unique among TRAFs in that it lacks a\nRING finger domain present in all other TRAFs and has the most \nrestricted expression pattern\; found almost exclusively in\nlymphocytes \, dendritic cells and certain epithelia. Overexpression\nof TRAF1 in many tumours of B cell origin is consistent with a\nsurvival function. I will discuss the role of TRAF1 in the survival\nof CD8 memory T cells. In addit ion\, the dependence of the 4-1BB\nreceptor\, a member of the TNFR family\ , on TRAF1 in mediating\nsurvival of CD8 T cells will be discussed. Using a systems biology\napproach novel TRAF1 binding partners have been identif ied to\nunderstand TRAF1-dependent-signalling pathways downstream of TNFRs \nin CD8 T cells.
