When host defense s are compromised\,\nbacterial killing by antibiotics is critical. Bacteri a can develop\nphenotypic multi-drug tolerance without harboring any inher itable\nmechanism of resistance\, and the mechanisms of tolerance remain\n largely unknown. Bacteria become tolerant under several growth\nconditions \, most notably when depleted for nutrients. We are\ninvestigating whether starvation confers tolerance by inducing a\nspecific response\, the strin gent response\, mediated by (p)ppGpp.\nNutrient starvation and induction o f the stringent response may\nalso be the common mechanism underlying the marked tolerance\nobserved in both stationary phase and biofilm growth. We studied\nthe opportunistic pathogen Pseudomonas aeruginosa to investigate \nwhether disruption of the stringent response impaired antibiotic\ntolera nce in-vitro and in-vivo\, and how (p)ppGpp mediated\ntolerance.
