A recently annotated human\ngenome sugges ts ca 20\,500 protein coding genes of which ca 8\,000\nhave no direct evid ence for a protein expressed and a further 5\,000\nwhere the evidence may be weak. Hence a human proteome project will\nprovide the missing evidence for protein expression by predicted\nprotein coding genes. Current effort s to define the proteome using\nantibodies monospecific to a representativ e protein for each\nprotein coding gene are ongoing with almost a third of the human\ngenome mapped in this way via the Human Protein Atlas. Current mass\nspectrometry efforts have enabled community standards to be\nimplem ented via test samples. The extension to label free methods\nto quantify p roteins by tandem mass spectrometry as well as their\nquantitation using a serial dilution of heavy isotope labelled\npeptides diagnostic of each re presentative protein provides the\nframework to define protein abundance f or each representative\nprotein in its major site of expression.
